Immunohistochemistry, Tissue Clearing and Confocal Imaging.

Several SPPIN research teams use optical and electrophysiological methods aimed at understanding brain functions. In order to complete these studies, I provide morphological data and analysis. This includes probing the expression of certain proteins (receptors, ligands, channels) using confocal microscopy and immunohistochemistry techniques, or tracing the connections between neurons expressing fluorescent proteins. These morphological descriptions, together with the data obtained by electrophysiology, help to construct coherent explanations for the phenomena observed by the researchers.

Main activities

Mapping the interactions between different brain regions by tracing signals of fluorescent proteins expressed in genetically defined neuronal cell-types labeled by stereotactically delivered adeno-associated viruses. To accomplish this I have developed tissue clearing techniques, three-dimensional imaging infrastructure and data processing to image through tissue sections of 1-3mm thickness and whole brains up to 8mm in thickness.

Morphological study of neurons: I develop and conduct immunohistochemistry techniques on brain slices to explore the expression of certain proteins (receptors, ligands, channels), or to study the networks formed by different brain cells.
